Generation of two CNR1 knock out/down colon carcinoma cell lines to assess GPR55 activity.

    Abstract

    Colorectal cancer is the third most commonly diagnosed cancer worldwide. Recent studies suggest that two receptors of the endocannabinoid system play an important role during the development of colorectal cancer, the cannabinoid receptor 1 (CB1 encoded by CNR1) and the G protein-coupled receptor 55 (GPR55), although very recent data also attest cannabinoid receptor 2 (CB2 encoded by CNR2) a role. While CB1 acts as a tumor suppressor by inhibiting cell proliferation, GPR55 is believed to be an oncogene, as it promotes cell proliferation, cell differentiation, cell division and migration indicating a role in metastasis. GPR55 is activated by its ligand LPI causing a phosphorylation of ERK and is inhibited by CB1 as long as CB1 is not activated. Both assumptions are partially proved by using GPR55 overexpressing colon cancer cell lines as well as mouse models with induced colon cancer. The interaction between CB1 and GPR55 in colon cancer cell lines was investigated.
